David Wigder, who has significant experience as an Environmental Engineer (2007) wrote that marketers have historically faced an uphill battle when it comes to marketing eco-friendly goods. Simply put, it is difficult to influence consumer purchase behavior without first impacting attitudes and values. These values, however, take a concerted effort over a long period of time to change. As a result, corporate marketers tend to stay clear of awareness and education communications, preferring to target consumers lower in the purchase-funnel who are already predisposed to green messaging. The reason for this is self-evident: when it comes to green, acquisition campaigns have higher and more immediate financial returns than awareness campaigns (Wigder, 2007). Yet, for marketers, the opportunity exists to influence environmentally friendly behavior without necessarily shifting attitudes. This effect has been subject of academic investigation including a study conducted by Professors John Thogersen and Folke Olander of the Aarhus School of Business (Denmark) examining the relationship between ââ¬Å"value prioritiesâ⬠and ââ¬Å"environmentally-friendly consumer behavior. â⬠(Wigder, 2007) As part of this study, Thogersen and Olander examined the impact of recycling on the values and behaviors of Danish consumers over the course of one year. (ââ¬Å"Human Values and the Emergence of a Sustainable Consumption Pattern: A Panel Study,â⬠Journal of Economic Psychology, 2002). The results of such investigation reveal several key findings that green marketers should consider: â⬠¢ First, the study reconfirmed that values drive behavior (while the converse relationship was not found to be statistically significant). While not surprising, this result confirms that marketers face an uphill battle if they are to influence environmentally friendly behavior without first addressing values. â⬠¢ Second, the study found that values are very stable and are difficult to impact in the ââ¬Å"short and medium term. â⬠â⬠¢ Finally, behavior change, the authors concluded, is hindered not only by values but by ââ¬Å"behavioral inertia, created by forces [such as established habits] that are independent of ââ¬â or at least not related in a simple way to ââ¬â valuesâ⬠. (Dobson, 2007) Yet significantly for marketers, the study also suggests that for those that already hold environmentally friendly values, environmentally friendly behavior can evolve over time if consumers are provided the opportunity to engage in this behavior. Thogersen and Olander concluded that ââ¬Å"when new opportunities for environmentally-friendly behaviour are offered, consumers holding ââ¬Ëenvironmentally-friendly valuesââ¬â¢ adjust their behaviour to be more consistent with their values. â⬠This finding implies that consumers who hold green values will demonstrate greener behavior if presented with relevant products or services (Wigder, 2007). Andrew Dobson wrote in an article called The Politics of Global Warming (2007), that in his review of the idea and practice of sustainable consumption, Tim Jackson points out that ââ¬Å"the rhetoric of ââ¬Ëconsumer sovereigntyââ¬â¢ and ââ¬Ëhands-offââ¬â¢ governance is inaccurate and unhelpfulâ⬠(see ââ¬Å"Motivating Sustainable Consumption,â⬠SDRN: Briefing 1). This is because consumption decisions take place within a cultural and institutional context which constitute the rules of the game, and which part determine the consumer decisions that people make. So when the iPod mini comes along hard on the heels of the only marginally larger original iPod, the social and economic context is geared to getting consumers to buy it (Dobson, 2007). In this context, as Jackson went on, ââ¬Å"policies based on information and price signals have had only limited success in changing unsustainable behaviorsâ⬠. Yet these are exactly the policies the government seems determined to pursue ââ¬â policies that, moreover, contribute to reproducing the pro-individual context that is part cause of our environmental problems. ââ¬Å"The dominant cultural model in 21st-century society is individualistâ⬠, wrote Tim Jackson. ââ¬Å"But this is only one form of social organization and there is evidence to suggest that it may not be sufficient to address the social complexity of pro-environmental behavioral change. â⬠But, policy-makers will say, policies based on price signals work with the grain of self-interest and are therefore realistic rather than aspirational as far as models of human motivation are concerned. Wrong. There is a growing body of social-science evidence to suggest that the self-interest model is actually a poor predictor of environmental attitudes and behavior (Dobson, 2007). For instance, in their survey of 4,000 individuals in four separate counties in Sweden, Simon Matti and Christer Berglund conclude that as far as pro-environment behavior is concerned, ââ¬Å"people are guided by other motives and values than the traditional economic rationality of the consumer â⬠¦ they feel a moral obligation to sort waste in order to contribute to a better environmentâ⬠(see ââ¬Å"Citizen and consumer: the dual role of individuals in environmental policyâ⬠, Environmental Policies, 15/4, 2006). More striking still, their research strongly suggests that policies designed to appeal to the individual as consumer rather than as citizen ââ¬Å"crowd outâ⬠, or reduce, ââ¬Å"the sense of moral obligationâ⬠in favor of pro-environmental activity. Once again, the preferred form of government policy both reinforces the frames of mind and conduct that contribute to environmental unsustainability and simultaneously undermines the habits and practices that inform much pro-environmental behavior. This double-whammy is a serious obstacle to dealing with climate change ââ¬â and indeed with any other problem which requires pro-social responses (Dobson, 2007). The fact that these results were garnered in Sweden may itself be significant. This is because a further piece of social-science research suggests that collectivist, social-welfare societies are a better incubator of pro-environmental behavior than individualist ones where welfare is looked on with suspicion. ââ¬Å"Those who place a high value on the welfare of others and on a collective approach to solving social problems are more likely to be willing to support environmental policies than those who do notâ⬠, writes finds Sharon Witherspoon (see ââ¬Å"Democracy, the environment and public opinion in Europeâ⬠, in W Lafferty & J Meadowcroft, eds. , Democracy and the Environment: problems and prospects (Edward Elgar, 1996). All of this suggests that addressing climate change is both more difficult and easier than the executive summaries swirling across the desks of government ministers and newspaper front-pages portray. It is more difficult, because the drivers of unsustainable attitudes and behavior are deeper and more structural than supporters of liberal capitalism can afford to believe. Yet it is also easier, because resistance to those drivers is expressed on a daily basis by the actions of tens of millions of citizens around the world as they strive to do the right thing, not for any gain for themselves or fear of fiscal punishment, but because itââ¬â¢s the right thing to do (Dobson, 2007). Governments assume that people donââ¬â¢t behave like that, and design policy accordingly. Social-science research suggests two things: first, that people do behave like this, and second, that government policy which fails to understand as much will not only be ineffective but ââ¬â in a move that converts tragedy into farce ââ¬â will undermine the very motivations for the behavior which it should be encouraging. Battle of York in the War of 1812 The Battle of York was fought April 27, 1813, during the War of 1812 (1812-1815). In 1813, American commanders around Lake Ontario elected to move against York (present-day Toronto), the capital of Upper Canada. Though lacking in strategic value, York presented an easier target than the main British base on the lake at Kingston. Landing on April 27, American forces were able to overwhelm Yorks defenders and captured the town, though promising young commander Brigadier General Zebulon Pike was lost in the process. In the wake of the battle, American troops looted and burned the town. Background In the wake of the failed campaigns of 1812, newly re-elected President James Madison was forced to reassess the strategic situation along the Canadian border. As a result, it was decided to focus American efforts for 1813 on achieving victory on Lake Ontario and the Niagara frontier. Success on this front also required control of the lake. To this end, Captain Isaac Chauncey had been dispatched to Sackets Harbor, NY in 1812 for the purpose of constructing a fleet on Lake Ontario. It was believed that victory in and around Lake Ontario would cut off Upper Canada and open the way for an attack on Montreal. In preparation for the main American push at Lake Ontario, Major General Henry Dearborn was ordered to position 3,000 men at Buffalo for a strike against Forts Erie and George as well as 4,000 men at Sackets Harbor. This second force was to attack Kingston at the upper outlet of the lake. Success on both fronts would sever the lake from Lake Erie and the St. Lawrence River. At Sackets Harbor, Chauncey had rapidly constructed a fleet that had wrested naval superiority away from the British. Meeting at Sackets Harbor, Dearborn and Chauncey began to have misgivings about the Kingston operation despite the fact that the objective was only thirty miles away. While Chauncey fretted about possible ice around Kingston, Dearborn was concerned about the size of the British garrison. Instead of striking at Kingston, the two commanders instead elected to conduct a raid against York, Ontario (present-day Toronto). Though of minimal strategic value, York was the capital of Upper Canada and Chauncey had intelligence that two brigs were under construction there. Landing with three more companies, Pike began forming his men when they were attacked by the grenadier company of the 8th Regiment of Foot. Outnumbering their attackers, who launched a bayonet charge, they repelled the assault and inflicted heavy losses. Reinforcing his command, Pike began advancing by platoons towards the town. His advance was supported by two 6-pdr guns while Chaunceys ships began a bombardment of the fort and Government House Battery. Directing his men to block the Americans, Sheaffe found that his forces were being steadily driven back. An attempt was made to rally around the Western Battery, but this position collapsed following the accidental detonation of the batterys travelling magazine. Falling back to a ravine near the fort, the British regulars joined with the militia to make a stand. Outnumbered on land and taking fire from the water, Sheaffes resolve gave way and he concluded that the battle was lost. Instructing the militia to make the best terms possible with the Americans, Sheaffe and regulars retreated east, burning the shipyard as they departed. As the withdrawal began, Captain Tito LeLià ¨vre was sent to blow up the forts magazine to prevent its capture. Unaware that the British were departing, Pike was preparing to assault the fort. He was approximately 200 yards away interrogating a prisoner when LeLià ¨vre detonated the magazine. In the resulting explosion, Pikes prisoner was killed instantly by debris while the general was mortally wounded in the head and shoulder. In addition, 38 Americans were killed and over 200 wounded. With Pike dead, Colonel Cromwell Pearce took command and re-formed the American forces. A Breakdown of Discipline Learning that the British wished to surrender, Pearce sent Lieutenant Colonel George Mitchell and Major William King to negotiate. As talks began, the Americans were annoyed at having to deal with the militia rather than Sheaffe and the situation worsened when it became clear that the shipyard was burning. As talks moved forward, the British wounded were gathered in the fort and largely left unattended as Sheaffe had taken the surgeons. That night the situation deteriorated with American soldiers vandalizing and looting the town, despite earlier orders from Pike to respect private property. In the days fighting, the American force lost 55 killed and 265 wounded, mostly as a result of the magazine explosion. British losses totaled 82 killed, 112 wounded, and 274 captured. The next day, Dearborn and Chauncey came ashore. After prolonged talks, a surrender agreement was produced on April 28 and the remaining British forces paroled. While war material was confiscated, Dearborn ordered the 21st Regiment into the town to maintain order. Searching the shipyard, Chaunceys sailors were able to refloat the aged schooner Duke of Gloucester, but were unable to salvage the sloop of war Sir Isaac Brock which had been under construction. Despite the ratification of the surrender terms, the situation in York did not improve and soldiers continued to loot private homes, as well as public buildings such as the town library and St. James Church. The situation came to a head when the Parliament buildings burned. Aftermath On April 30, Dearborn returned control to the local authorities and ordered his men to re-embark. Before doing so, he ordered other government and military buildings in the town, including the Governors Residence, deliberately burned. Though a victory for American forces, the attack on York cost them a promising commander and did little to alter the strategic situation on Lake Ontario. In fear, animal is more favorable to dark area which was shows in negative control. Force Swim Test (FST) Time cycle in seconds was count in all groups. Time cycle per five minute were significantly increased (P>0.001) in Semecarpus anacardium at dose of 200 mg/kg (20à ±4.2044) compared with negative control (25à ±2.5421). Light and Dark Test After immobilization of animals for 3 hr, the drug treatment was started for all groups except negative control. Time spent in light and dark area was observed. Time spent in light area were significantly increased (P>0.001) after administration of Semecarpus anacardium at dose of 200mg/kg (178à ±3.5041 sec.) as compared with negative control (58à ±2.1245 sec.). In fear, animal is more favorable to dark area which was shows in negative control. Open Field Test (OFT) OFT is the test to evaluate anti- anxiety effect as well as to compare the statistics with actophotometer because each squire in OFT is 10 Ãâ"10 cm and each electrodeââ¬â¢s difference in actophotometer is 6 cm so the reading should be double in OFT. Animal in control group were shows significant walk fullness in OFT (45à ±2.2405 sec.). After administration of Semecarpus anacardium at dose of 200mg/kg, the animal was shows significant effect (P>0.001). Rearing is the parameter in OFT which shows alertness of animals. The gaseous messenger molecule nitric oxide (NO) is synthesized from its precursor L-arginine by a family of three NO Synthases (NOS), designated as ââ¬Å"neuronalâ⬠NOS-I, ââ¬Å"inducibleâ⬠NOS-II and ââ¬Å"endothelialâ⬠NOS-III. In the adult brain, the inducible iso form NOS-II is present only at very low levels in microglia and immune cells, while ââ¬Å"endothelialâ⬠NOS-III is expressed predominantly in the vasculature. Whether or not this isoform is also expressed in neural cells, is still a matter of debate but data arguing for this are only sparse. The quantitatively major source for NO in the CNS thus is the ââ¬Å"neuronalâ⬠isoform NOS-I present in approximately 1% of all neurons. Nitrinergic transmission is especially important in limbic structures, in the basal ganglia where NO regulates striatal output and in the cerebellum. NO exerts multiple action in the CNS and from animal studies, it has been suggested that it is involved in behavioral p rocesses such as learning and memory formation. Pathologies of the NO pathway have been implicated in almost every major neuropsychiatric disorder including Schizophrenia, affective disorders, Alcoholism, Alzheimerââ¬â¢s dementia, Parkinson and Huntingtonââ¬â¢s disease. For some of these disorders, NOS-I has also been identified as a risk gene in human case-control association studies. The role of NO in the regulation of normal human brain functioning however is still unclear, although first genetic studies argue for a function of NOS-I in the regulation of impulsive behaviors. In a second series of experiments, we investigated whether NOS-I knockdown animals have cognitive deficits. Plasma nitrates level was significantly decreased (P>0.001) after administration of Semecarpus anacardium at dose of 200 mg/kg (52.23à ±2.1401sec.) as compared with negative control (74.24à ±2.2406). In fear or anxiety, animal were showed increased level of plasma nitrates which was shows in negative control. iNOS level was significantly increased (P>0.001) after administration of Semecarpus anacardium at dose of 200mg/kg (78.37à ±3.2131sec.) as compared with negative control (26.23à ±2.5470 sec.). In addition to its role in cholinergic transmission, substantial evidence has accumulated over the last two decades which suggests a non- cholinergic neuromodulatory function for AChE. Few studies have demonstrated that the expression of AChE during early development correlate closely with the major phase of neurite outgrowth. Layer et al. have showed that AChE inhibitors have been shown to retard neuritic outgrowth in a dose dependent manner in retinal ganglion cells, dorsal root ganglion and sympathetic ganglion neurons. There is a growing body of evidence supporting the morphogenic effects of AChE in both in vivo and in vitro systems. AChE is known to regulate the neuritic outgrowth and survival of cultured neurons and also has morphogenic and axogenic role in the developing nervous system. In addition, AChE has a role in cell growth and survival. These functions are considered to be the non-classical roles of this classical enzyme. Furthermore, ACh is also known to enhance the ne uritic outgrowth and in turning of the nerve growth cones. These studies, together with the present demonstration of increased dendritic arborization in the hippocampus, suggest that chronic drug administration induces AChE activity which in turn might modulate dendritic branching pattern in specific brain regions. Ach level was significantly decreased (P>0.001) after administration of Semecarpus anacardium at dose of 200mg/kg (53.26à ±2.0987 sec.) as compared with negative control (81.23à ±3.0245 sec.). The efficacy of this plant extract toward the transmitters was significant. MAO regulates metabolic degradation of catecholamine, serotonin and other endogenous amines in CNS. Inhibition of this enzyme causes reduction of metabolism of these transmitters and subsequent increase of these biogenic amines. MAO-A level was significantly decreased (P>0.001) after administration of Semecarpus anacardium at dose of 200mg/kg (56.6à ±3.3245 sec.) as compared with negative control (86.1à ±2.3024 sec.). MAO-B level was significantly decreased (P>0.001) after administration of Semecarpus anacardium at dose of 200 mg/kg (44.8à ±3.2431 sec.) as compared with negative control (73.4à ±2.2061 sec.). Glutathione reductase level was significantly decreased (P>0.001) after administration of Semecarpus anacardium at dose of 200mg/kg (1478.5à ±3.2436 sec.) as compared with negative control (1634à ±2.2102 sec.). All values are expressed in U/I. Glutathione reductase level was decreased after administration of extract of Semecarpus anacardium at dose 200 mg/kg in mice. Glutathione reductase is the enzyme which increases in anxiety and depression. This enzyme secretes from hippocampus region of brain. The level of this enzyme was significantly reduced in mice compared with vehicle treated control group. On the bases of behavioral as well as biochemical estimation, study concludes that Semecarpus anacardium shows significant effect in plasma nitrates and other chemical messenger in anxiety at dose of 200mg/kg compared with negative control. 9. SUMMARY AND CONCLUSION The present study is designed to evaluate ââ¬Å"Effect of Semecarpus anacardium on plasma nitrates on stress induced anxiety in miceâ⬠. Behavioral parameters show following result: After administration of Semecarpus anacardium Time spent in open arm in Elevated Plus Maze, Time cycle per five minute in Force Swim Test, Time spent in light area in Light and Dark Test, No. of Squire Cross in Open Field Test was significantly increased after administration of Semecarpus anacardium at dose of 200 mg/kg as compared with negative control. Biochemical Estimations show following result: Plasma nitrates level, Ach level, MAO-A level, MAO-B level, Glutathione reductase level was significantly decreased after administration of Semecarpus anacardium at dose of 200 mg/kg as compared with negative control. iNOS level was significantly increased after administration of Semecarpus anacardium at dose of 200mg/kg as compared with negative control. On the bases of behavioral as well as biochemical estimation, study concludes that Semecarpus anacardium shows significant effect in plasma nitrates and other chemical messenger in anxiety at dose of 200mg/kg compared with negative control. 6. Materials Methods 6.1 Materials Collection Authentication The plant Semecarpus anacardium has been taken from local market authenticated from Department of Botany Dr. H.S. Gour University, Sagar M.P. Herbarium No. Bot./her/A/1124. Extraction procedure 6.3.1 Petroleum ether extract: The whole plant nuts was cleaned and shaded dried for 10-15 days. The dried nuts were pulverized by an electrical blender and nut paste obtained. About 30-40 g of the nut paste was subject for extraction with 400 ml of Petroleum ether solvent by Soxhlet apparatus for 24 hrs. Constant heats of 50 60 0C provided by Mantox heater of Soxhlet for recycling the solvent. The extract was concentrate using Rotary evaporator at 60 0C for 20 min at a speed of 5m/s. The concentrated extract kept in refrigerator at 4 0C for further use. (50) 6.3.2 Ethanol extract: The nuts were shed dried for about 20 days and then subsequent to reduce coarse drug particle into fine powder using pestle and mortar. The extraction was carrying out by ethanol solvent Soxhlet extraction techniques. Solvent used consecutively with gradient polarity. The extract evaporated to complete dryness by using vacuum distillation and kept in refrigerator for further use. (51) Phytochemical screening 6.4.1 Tests for Alkaloids Mayerââ¬â¢s Test: Extract treated with Mayerââ¬â¢s reagent (Potassium Mercuric Iodide). Formation of a yellow coloured precipitate indicated the presence of alkaloids. Wagnerââ¬â¢s Test: Extract treated with Wagnerââ¬â¢s reagent (Iodine in Potassium Iodide). Formation of brown/reddish precipitate indicated the presence of alkaloids. Dragendroffââ¬â¢s Test: Extract treated with Dragendroffââ¬â¢s reagent (solution of Potassium Bismuth Iodide). Formation of red precipitate indicated the presence of alkaloids. Hagerââ¬â¢s Test: Extract treated with Hagerââ¬â¢s reagent (saturated picric acid solution). Presence of alkaloids confirm by the formation of yellow coloured precipitate. Tannic acid test: Extract treated with 10% Tannic acid solution. Alkaloids gave buff colour precipitate. (52) 6.4.2 Detection of Phenols Bromine water test: Test solution treated with few milliliters of bromine water. Formation of yellow precipitate indicated presence of Phenols. Ferric chloride test: Test solution gave blue green colour with ferric chloride. (53) 6.4.3 Detection of Saponins Emulsion test: 1 ml of the extract filtrate added to few drops of olive oil. The mixture added to another two drops of olive. The mixture shakes and observed for the formation of emulsion. Frothing test: 1 ml of the extract filtrate diluted with 4 ml of distilled water. The mixture was shake vigorously and then observed on standing for a stable froth. 6.4.4 Detection Steroids and Triterepenoids Libermann- Buchard test: Extract treated with few drops of acetic anhydride, boil and cool, conc. Sulphuric acid added from the sides of the test tube. Formation of a brown ring at the junction of two layers and the upper layer turns green which shows the presence of Steroids and formation of deep red colour indicated the presence of Triterepenoids. Salkowski test: Treated extract in Chloroform with few drops of cone. Sulphuric acid, shaked well and allowed standing for some time, red colour appeared at the lower layer indicates the presence of Steroids and formation of yellow coloured lower layer indicated the presence of Triterepenoids. 6.4.5 Detection of Tannins Lead sub-acetate test: 1 ml of the filtrate added to 3 drops of the lead sub-acetate solution. A cream gelatinous precipitate indicated the presence of tannins. Ferric chloride test: 1 ml of the filtrate diluted with distilled water and added with 2 drops of ferric chloride. A transient greenish to black colour indicated the presence of tannins. 6.4.6 Detection of Flavonoids Shinoda test (Magnesium Hydrochloride reduction test): To the test Solution, added few fragments of Magnesium ribbon and added concentrate Hydrochloric acid drop wise, pink scarlet, crimson red or occasionally green to blue colour appeared after few minutes. Alkaline reagent test: To the test solution added few drops of sodium hydroxide solution; formation of an intense yellow colour, which turned to Colourless on addition of few drops of dil. acid, indicated presence of Flavonoids. Ammonium test: A quantity (4 ml) each of the filtrates was shaking with 1 ml of dilute ammonia solution (1%). The layers allowed to separating. A yellow coloration at the ammonia layer indicates the presence of Flavonoids. Aluminium chloride test: A quantity (4 ml) each of the filtrates was shake with 1 ml of 1% aluminium chloride solution and observed for light yellow coloration. A yellow precipitate indicated the presence of Flavonoids. 6.4.7 Detection of Anthraquinones 1. Dilute sulphuric acid (5 ml) added to 0.1 g of the test extract in a test tube and boil for 15 min in a water bath. It was then cool and neutralize with 20% potassium hydroxide solution. A mixture, 10 ml of equal parts of Fehlingââ¬â¢s solution A and B will add and boil for 5 min. A more dense red precipitate indicated the presence of glycoside. 2. About 0.5 ml of extract taken and subject to the following tests.1 ml of glacial acetic acid containing traces of ferric chloride and 1ml of concentrate sulphuric acid added to the extract and observed for the formation of the reddish brown colouration at the junction of two layers and the upper layer turned bluish green showed presence of Glycosides. Pharmacological Screening 6.5.1 Animal: Mice required as Animal model Body weight: 25 gms. Floor area per animal: 15 in2. Cage height: 5 inch. Temperature: 64à ° to 79à °F (18à ° to 26à °C). Relative Humidity: 40% to 70%. Number of air changes per hour: 10 ââ¬â 15. Light levels: 30 foot-candles. Duration of Light: 12 -14 hours. Duration of Darkness: 10 12 hours. 6.5.3 Biochemical Estimation 6.5.3.1 Plasma Nitrate estimation: Plasma nitrate were measured by spectrophotomeric assay based on Griess reaction. Blood were withdrawn from tail vein of mice and plasma were using cooling centrifuge at 2500 rpm for 10 min. Plasma were mixed with equal volumes of Griess reagent (1% Sulphanilamide+ 0.1% naphthylelediamine dihydrochloride+ 2.5 % phosphoric acid) and incubated at room temp for 10 min. to yield a chromophore. Absorbance was read at 543 nm spectophotometrically.(59) 6.5.3.2 i NOS estimation: Sample collection After the behavioral tests, three mice from each group was deeply anesthetized and perfuse with 4% paraformaldehyde for subsequent Nissl staining. The other animals were anesthetized and kill; blood was collected and brains were removed. Blood, anticoagulated with 1.5% EDTA centrifuged at 12,000 rpm for 10 minutes, and then the supernatant was collected. All these samples stored at âËâ80à °C for further analysis. RNA extraction and reverse transcription Total RNA extracted from the brain tissue using Trizol reagent. Total mRNA (1 à ¼g) was transcribed using Quant script cDNA RT Kits according to the manufacturerââ¬â¢s manual. Briefly, RNA (1 à ¼g) pretreated with DNA-free DNase treatment and removal reagents. RNA samples incubated with a mixture consisting of containing dNTPs, random primers, 10Ãâ" RT mix, Quant Reverse Transcriptase, a reverse transcriptase and RNase-free water to a final volume of 10 à ¼l at 37à °C for 1 h. Real-time RT-PCR cDNA l used for quantification of mRNA by real-time RT-PCR. Real-time RT-PCR will perform on an Applied Rotor-Gene 3000 under the following conditions: iNOS and GAPDH for 40 cycles at 94à °C for 30 s, 63à °C for 60 s, and 72à °C for 90 s. Relative quantitative measurements of target gene levels was performed using the ÃâCt method, where Ct is the threshold concentration. The Treatment of Men and Women in Tess of the D'Urbervilles Tess now feels he can understand her problem. When she tells him he refuses to accept it, he has placed Tess on a pedestal and is mortified when she falls from it. She begs his forgiveness but he has become very cold towards her and doesn't seem to hear her pleas. Hardy really shows the double standards of the day by making both crimes so similar. We can see how differently men and women were treated. Today both genders are treated the same and Tess is seen as a victim, Angel gave consent and so is now seen as the one in the wrong. Angel is very cold and refers to Tess as a different person not the woman he loved. 'You were one person, now you are another'. Angel is too rational and socially aware of Tess' disgrace to forgive her. Tess is so distraught he cannot forgive her she contemplates taking her life but she can't because it will bring suspicion and shame on Angel. There is a Chinese proverb that says, ââ¬Å"One chopstick is easily broken, but ten pillars chopsticks firmly hold dough.â⬠This reveals a simple rule that unity is much stronger than individuals, and if you do not want to be beaten, you should unite and cooperate against enemies together. This proverb can also be used internationally. Global cooperation is important to maintain economy, and improve safety, peace and the environment. Different countriesââ¬â¢ economies are linked together, and influence one another in many ways. Hence, in the globalized economies people always worry more about the economic growth and crisis. Trade is the activity of exchanging goods and services. There are many trades, for example, to exchange fish for beef or to pay a taxi for its driving. One of the economic principles is that trade makes everyone better off. International trade is essential in these days because each country cannot provide the product it needs to serve the whole society, or it will cost more opportunity cost such as time and money in some productions. In this case, global trade is required. According to the World Trade Organization (WTO), 2013 annual exports valued $18,784,000 million in total merchandise, and valued $4,623,710 million in commercial services (2014). These massive amounts of numbers show the significant value of international trade in the world. China is the largest exporter, and had valued $2,209,626.0 million, which is 11.7 percent of total world merchandises trade in 2013. Also, it had become the world second large economy according to WTO Database. However, it was only exporting $5,790 million goods and 1.0 percent of total world trade in 1973 (WTO, 2014). China has an unbelievable growth, and the biggest reason is that China started to switch the closed economy into more tradable market economy and trade much freely with other countries since 1978 (Chinese economic reform). The international trade provides more job opportunities, attracts investments and brings advanced technology, and these are the basic elementsà in order to gain economic growth. Unfortunately, the drawback of collapsing a big international bank Lehman Brothers is unimaginable ââ¬â crash the global financial system, credit crunch, and economic regression. Because of the Asian Crisis in 1997, the Association of Southeast Asia Nations (ASEAN) had been more careful against crises such as no risky loans and strengthened nations cooperation. China decided to set up a $ 10 billion total fund in order to support China-ASEAN work and offer $39.7 million aid to help Cambodia, Laos and Myanmar in financial troubles (Yan, W. 2009). International cooperation is needed for peopleââ¬â¢s safety such as natural disasters. Disasters are extremely powerful and destroy millions of lives. According to America, the Indonesia tsunami, which is caused by a 9.0 magnitude earthquake in the sea near Banda Aceh in 2004 had killed over 150 hundred of people and created millions of homeless among Southern Asia countries and Africa. However, the number was continuously increasing due to injury and disease. Governments provided about $2 billion to support those countries in the disaster. This was a global cooperation ââ¬â many organizations such as Catholic Relief Services, the Red Cross, Oxfam, Caritas and Doctors Without Borders helped to save lives from diseases and water pollution. Because the transportations are damaged the supplies were delivered by helicopters and ships, which were supported by United State Military. Meanwhile, individualsââ¬â¢ donations around the world were greater than the governmentsââ¬â¢ total amounts, and these would be used in future recovery (2005). These international saving was successful and helped for countless people. Peace is a big issue for people because there are still numerous shouting and wars going on. In addition, the most thing that cause these dangerous issues are weapons. Weapons are made to protect people, however, they have become more and more powerful especially the nuclear weapons. The total nuclear weapons in the world are enough to destroy the earth! Also the negative effect of nuclear bombs not only it can destroy a city immediately but also spread the radiation, which has decades, even different generation influences after exposure. Since 1945, the statistics show that the total war and violent has diminished. However, this kind of weapon is still being produced in many nations. Safety of citizens is questing because of threats of terrorisms. They can buy or make nuclear weapons and expose ââ¬Å"dirty bombsâ⬠(Lifton, R. 2013). In this case, terroristsà should be stopped; people should be protected and no nuclear weaponsââ¬â¢ threat. Non-nuclear weapons are very important and hard to achieve. In addition, it needs global cooperation ââ¬â every government does their duty to increase the world level of peace. Most importantly, the global environment affects all lives on the earth, and countries should avoid ruining it. First is the global warming issue, it is a big issue that is caused by human activities and has the additional effect of climate change. In 2009, United Nations Climate Change Conference in Copenhagen had failed to make a worldwide agreement about emissions reduction (Nordhaus, D. 2011). This is because the disagreement about the distribution of reducing greenhouse gases and its costs. The decrease in temperature is essential because it would affect everyone on the earth. Nevertheless, without governmentsââ¬â¢ cooperation this goal cannot be achieved efficiently because different places and regions have different situations and meet variable difficulties. For example, if the cost for imposing a high carbon dioxide emissions tax will generate the decline of Gross Domestic Product (GDP), and the influence of economy may even promotes a financial crisis, a government will not choose to do that. Also, when a country with smaller population but higher per person green house gas emission but another nation which has the opposite situation, their will be conflict when ask them to produce the same amount of emission. These are complicated and hard to negotiate an agreement. However, as the future cost for climate change is too high ââ¬â decrease in generalââ¬â¢s life span and quality, international cooperation cannot stop and require to be done more successfully. Besides, climate change also affects other organisms such as animals and plants. Human activities such as travelling may cause many animals and plants in danger. Decrease in animals and plants will damage the ecological chain even influence the whole planet. To protect those animals and plants, the Washington Conversation has established in1975, July 1st and called the Convention on International Trade in Endangered Species of Wild Fauna and Flora, and it goal is to ensure the safety of wild species not dangerous because of international trade (CITES). Because the trade is worldwide and every country will affect neighboring countries, CITES needs to make sure every country does their work to stop threat other species in other countries. In general, many things include economy, human lives and other species are interacting withà each individual and country. They will promote different but widespread effects. Because of these facts it needs global cooperation to enhance the quality and safety in these fields. Governments should keep improving international action in order to provide a better world for all of lives.
